This might appear contradictory since after cocaine exposure (which induced increase of opioid receptors expression) at 24 hpf, we expect a reduction in dre-let-7d-1. This could be due to the fact that cocaine has different actions in both precursors dre-let-7d-1 and dre-let-7d-2 (Fig. 2B and C, respectively) as well as in the mature miRNA dre-let-7d (Fig. 2A). According to ISH, the expression of dre-let-7d was increased in embryos exposed to cocaine at 24 hpf in the telencephalon, midbrain, MHB and hindbrain (Fig. 10B, D and F), and at 48 hpf cocaine upregulated the expression of dre-let-7d in the telencephalon and MFL (Fig. 10H, J and L). [score:8]
The silencing and overexpression of opioid receptors affected the expression of dre-let and its precursors, and the similar expression of opioid receptors and let-7d miRNA in the CNS and at the periphery indicates that they have a close relationship. [score:7]

The expression of the dre-let-7d in the CNS (Fig. 4) suggest that this miRNA has an important role during the formation of the CNS (24 hpf) and primary organogenesis (48 hpf) in zebrafish embryos. [score:3]

Surprisingly, the expression level of let-7 miRNA in lin-28a and lin-28b morphant embryos was very low, and we did not detect significant upregulation in these morphants, suggesting that the downregulation of Lin-28a or Lin-28b is itself insufficient to induce the expression of the let-7 family, at least at 5 hpf (Figure 5B). In a reciprocal fashion, Lin-4 and let-7 expressions were first detected at L1 and L3 stage, respectively and became stronger as development proceeded, thereby down-regulated the expression of their target gene. [score:11]
Since zebrafish lin-28b and lin-41 have a lin-4/miR-125b and let-7 target sites in their 3′UTRs (using Targetscan and PITA) [33], the downregulation of lin-28b and lin-41 might be directly regulated by these miRNAs. [score:10]

Since LIN-28 prevents the premature accumulation of let-7 miRNA in L2, let-7 miRNA expression occurs during the third larval stage (L3) and controls the fourth larval stage (L4)-to-adult transition by repressing multiple target genes, including the TRIM protein Lin-41 [6], [18], [19]. [score:5]
We also observed the expression of lin-41, a down-stream target of let-7 in the presumptive anterior neural plate. [score:5]
Consistent with a prominent increase in let-7 miRNA, RT-PCR analysis of lin-41 expression showed that lin-41 mRNA was downregulated in both lin-28a and lin-28b morphants as compared to the control sample at 24 hpf (Figure 5D). [score:5]

Intrigued by our results on the analysis of miR-24 and let-7 mutations, we reasoned that a comprehensive analysis of our mutagenized miRNA sequences would establish the most successful mutations to functionally perturb miRNA expression in vivo (Fig. 4A). [score:5]
